Joe joined the Boston Celtics as an assistant coach in June 2019. Ime Udoka's suspension for the 2022–23 season was announced on September 22, 2022, and he was named the Celtics' interim head coach. Following the Celtics' league-best 18-4 start to the season, he was named Eastern Conference Coach of the Month for October and November on December 1, 2022.

He was appointed as the head of Team Giannis for the 2023 NBA All-Star Game on January 30, 2023. Mazulla became Celtics' main coach on February 16, and his contract was extended.

Joe Mazzulla played as the University of West Virginia's backup guard from 2006 to 2011. He withdrew in 2008-09 and later joined as a junior captain for the 2009-10 season and led the Mountaineers to the Final Four. He later became the senior captain in 2010–2011 and averaged 7.7 points, 3.8 rebounds, and 4.2 assists per game.

He is an American of Italian descent. His parents are Daniel Mazzulla Jr. and Latresa Mazzulla. Joe grew up in Johnston with two younger siblings, Justin and Gianna.

Joe Mazzulla's father was also a basketball coach who played college basketball at Bryant and also played professional games for five years in Chile. Dan Mazzulla was inducted into the Bryant Hall of Fame in 2007. He succumbed to cancer in April 2020.

He is married to Camai Mazzulla. They met while playing basketball at West Virginia University, where he was a student. They tied the knot in 2014 after graduating and have been together ever since.

Joe worked at West Virginia as an assistant coach for three seasons after graduating. The NBA G League hired him as an assistant coach after that.

He could not help but mention how his time as a student helped him meet his wife when he left West Virginia. Joe Mazzulla wrote about their relationship and the time they spent developing as a couple while in college and after, saying:

"I have spent some of my most significant years here, including my college years and the years after graduation when I continued to develop emotionally and spiritually. I also had the great fortune of meeting my lovely wife, getting married, starting our family here, and buying our first house. My family and I will always be grateful for the experiences West Virginia has provided us."

Camai is the mother of two boys, Michael Harden and Emmanuel, who she characterised on the "More Than a Season" podcast as "two very wild and crazy boys." The Boston Celtics' coach relocated with his wife and kids to Boston to maintain their family's unity when he accepted an assistant coaching position in 2019.

The wife of Joe Mazzulla is no stranger to sports; during her time in college, she competed in both volleyball and track. She started leading the school's volleyball team after earning her degree from Glenville State College. She worked there as the head coach from 2010 to the 2013 season, and she revealed that she decided to stop because she had "made a commitment at that point to invest in her husband's career."
