The 23-year-old star rose highest to convert a corner by Lorenzo Insigne in the 15th minute to double Napoli's lead.
Osimhen also provided the third goal for Hirving Lozano in the 19th minute.
Chukwueze grabs consolation for Villareal
Samuel Chukuweze was on target for Villarreal as they suffered a 2-1 loss to Alaves.
Unai Emery made a lot of changes from the midweek loss to Liverpool in the Champions League as Villarreal found themselves two goals down at halftime.
But just after the restart Chukwueze pulled one back after a nice solo run and a terrible mistake by Alaves goalkeeper. But the Yellow Submarines could not get the much-needed equalizer.
