After a barren first half, the host showed no signs of exertion from midweek as they controlled most of the second half.
With 18 minutes remaining, red-hot forward Jarrod Bowen opened the scoring for the Hammers.
Six minutes later, Kudus doubled the advantage after a quick one-two with Lucas Paqueta before firing through the legs of Johnny Evans, beating goalkeeper Andre Onana.
In a video shared on social media, Kudus had won the ball in midfield before giving it to Paqueta, who laid it back to the Ghanaian for West Ham's second.
The former Ajax midfielder has now netted nine goals since joining the London outfit, with the strike against United his sixth in the Premier League.
Kudus was playing for the first time against his former manager Erik ten Hag.
